灵芝孢子粉对2型糖尿病大鼠睾丸损伤活性氧机制的探讨 被引量:6

摘要 目的:探讨灵芝孢子粉对2型糖尿病大鼠睾丸损害的活性氧(RO S)机制。方法:W istar雄性大鼠40只,随机分组。模型组及灵芝组大鼠经尾静脉一次性按25 m g/kg注射2%链尿佐菌素(STZ),正常对照组尾静脉注射等量柠檬酸钠-柠檬酸缓冲液。正常饮食喂养2周后,进行糖耐量试验,模型组和灵芝组以糖耐量异常者保留,余去除,并改喂高脂高糖饮食,灵芝组另加灵芝孢子粉250 m g/kg.d持续10周,实验结束前1d做糖耐量试验,取双侧睾丸检测组织中T水平、M DA含量和SOD、G SH-Px活性。结果:糖尿病组与正常对照组比较,T水平、SOD、G SH-Px活性降低,差异有显著性意义(P<0.01),M DA含量明显高于对照组(P<0.01)。而灵芝孢子组与糖尿病组比较,T水平、SOD、G SH-Px活性升高,差异有显著性意义(P<0.01),M DA含量明显低于糖尿病组(P<0.01)。结论:糖尿病RO S增多,抗氧化酶(SOD,G SH-Px)活性下降,可能是造成睾丸细胞损伤,导致睾丸功能紊乱,进而导致性与生殖功能障碍的原因。而灵芝孢子粉能有效降低M DA的含量对糖尿病大鼠生殖系统的毒性损害,增强SOD、G SH-Px活性,可能是其对糖尿病大鼠生殖系统的细胞凋亡有明显抑制,从而对睾丸细胞起保护作用的机制之一。灵芝孢子粉作为一种治疗糖尿病的辅助用药,在临床应用上具有广阔前景。 Objective: To observe and approach the influence of Ganoderma lucidum spore on the mechanism of ROS damage to NIDDM rat testis. Methods: Fifty male Wistar rats were divided randomly into three groups (normal group, model group and ganoderma group). The model and ganoderma group rats were injected 2% STZ once through vena caudalis, and the normal group rats were injected tales-doses sodium citrate - citrate buffer solution. After normal diet had been fed for 2 weeks, the progressed glucose tolerance test was carried out, and abnormal glucose tolerance rats in the model and ganoderma group were retained and then given high lipid and carbohydrate food and drink, and at the same time the spores of Ganoderma lucidum (250mg/kg · d) was added to the ganoderma group for 10 weeks. The progressed glucose tolerance test was performed one day before the experiment was completed, and the rats were decapitated for the measurement of the correlated indexes Results: The NIDDM model was prepared succeedly , and compared with normal group, the T level and GSH-Px and SOD activities dropped sharply with the significant difference (P 〈 0.01) and the MDA content obviously increased (P 〈 0. 01) in NIDDM model group. Compared with model group, the T level and GSH-Px and SOD activities dropped sharply with the significant difference (P 〈 0.01) but the MDA content obviously decreased (P 〈 0.01) in ganoderma group. Conclusion:Increased ROS and decreased antioxidant enzymes (SOD and GSH -Px) activities might give raise to testis damage and its dysfunction , resulting in sexual and reproductive malfunction. Ganoderma lucidum spore can reduce MDA content and increase the activities of GSH-Px and SOD, and this may be one mechanism by which Ganoderma lucidum spore restrains the apoptosis of generate cells caused by diabetes to prevent the damage to generate cells. Ganoderma lucidum spore is an assistant medicine to cure diabetes, useful in clinic application.
