摘要 目的:观察银杏叶提取物(EGB)对大鼠海马神经干细胞(N SCS)增殖及分化为星形胶质样细胞的影响。方法:取新生24h内的W istar大鼠海马组织的细胞进行体外培养,1周后将其接种在培养板中,观察不同浓度的EGB对细胞的生长作用,并检测胶质原纤维酸性蛋白(GFAP)的表达。结果:各实验组N SCS的生长明显好于对照组,免疫细胞化学显示:同一时间内,诱导N SCS分化为星形胶质样细胞的百分率增高;同一浓度的EGB,诱导神经干细胞分化为星形胶质样细胞的百分率随着时间的延长呈上升的趋势。结论:EGB能促进神经干细胞的存活、增殖及向星型胶质样细胞的方向分化。 Objective: To observe the proliferative and differentiative effects of extracts of ginkgo biloba leaves (EGB) on rat Hippocampal neural stem cells(NSCs). Methods: Hippocampal NSCs from Wistar rats born within 24 h were cultivated in vitro. After a week ,these cells were equally plated into three pieces of 24 well plates, and each plate was equally divided 4 groups. At some moment , we observed the growth and detected the proliferation and the GFAP expression(astrocyte-special marker). Results: The growth of every experimental group was better than that of control group. The percentage of astrocyte-like rose with increasing EGB concentration, and the percent of the astrocyte-like rose with time delay at the same concentration of EGB. Conclusion: EGB can promote the survival,proliferation and differentiation of NSCs, especially the differentiation of NSCsinto the astrocyte-like.
