摘要
目的:建立以高效液相色谱法测定人血浆中阿糖胞苷浓度的方法。方法:色谱柱为Zorbax C18,检测波长为280nm,流动相为0.01mol/L磷酸盐(pH=7.0)-乙腈(95:5),流速为1.0ml/min。结果:阿糖胞苷血药浓度在0.05~20μg/ml(r=0.9999)范围内线性关系良好,最低检测浓度为2ng/ml,方法回收率为98.3%~103.2%,日内、日间相对标准差分别为2.8%、3.6%。结论:本方法简单、快速、准确,适用于阿糖胞苷临床血药浓度监测和药动学研究。
OBJECTIVE:To establish a HPLC method for the determination of cytarabine in human plasma. METHODS: The assaying was performed on Zorbax-C18 column,the mobile phase consisted of 0.01mol/L sodium phosphate (pH = 7.0) - acetonitrile(9S : S)with flow rate at 1.0ml/min and UV detection wavelength at 280nm .RESULTS: The linear blood concentration range for cytarabine was 0.05~20μg/ml( r = 0.9 999)with the minimum detection limit at 2ng/ml, the method recovery was 98.3% ~ 103.2% , the relative standard deviations of intra - day and inter day were 2.8% and 3.6% , respectively.CONCLUSIONS:This method was simple, fast and accurate,and suitable for the assaying of cytarabine in human plasma and pharmacokinetics study.
