摘要
目的观察低剂量十一酸睾酮(TU)对心肌梗死后慢性心力衰竭雄性大鼠血清细胞因子的影响。方法120只健康雄性SD大鼠随机行冠状动脉结扎(105只)或假手术(15只)。6周后将存活模型鼠随机分为2组:心力衰竭+TU治疗组(22例)给予TU5mg/(kg·2w)肌肉注射;心力衰竭组(22例)给予安慰剂。用药12周后测定血清睾酮(T)及肿瘤坏死因子α(TNFα)和白细胞介素10(IL10)水平。结果心力衰竭组雄鼠血清T水平明显低于假手术组(153.91±77.45与475.28±161.37)ng/dl,P<0.05,心力衰竭+TU组T浓度与假手术组差异无显著性(P>0.05)。心力衰竭组较假手术组血清TNFα水平明显升高(8.59±3.22与5.29±2.30)ng/L,P<0.05,IL10明显下降(4.90±1.32与7.30±2.14)ng/L,P<0.05。心力衰竭+TU组较心力衰竭组血清TNFα水平显著下降(5.97±1.81与8.59±3.22)ng/L,P<0.05,IL10显著升高(6.55±2.11与4.90±1.32)ng/L,P<0.05。心力衰竭+TU组与假手术组血清TNFα和IL10水平差异均无显著性(P>0.05)。结论低剂量睾酮可降低心肌梗死后慢性心力衰竭雄性大鼠血清TNFα水平,提高IL10水平。
Objective To investigate the effects of low dose testosterone undecanonate (TU) on serum cytokines in male rats with postinfarction chronic heart failure (CHF) model. Methods 120 SD male rats were randomly assigned to ligation of coronary artery ( n = 105 ) or pseudo operation ( PS, n = 15 ) . 6 weeks later,44 survived model rats were randomly divided into 2 groups: CHF + TU group( n =22) treated with TU 5 mg/kg per 2 weeks intramuscular injection and CHF group( n =22) treated with placebo. Sermn testosterone( T) and Tumor necrosis factor-α(TNF-α) and IL-10 levels were determined after treatment of 12 weeks. Results Serum T levels in CHF group were significantly lower than those in the PS group[ ( 153.91 ± 77.45 ) ng/dl vs (475.28 ± 161.37 ) ng/dl, P 〈 0.05 ], serum T level in CHF + TU group was not significantly changed compared with that in PS group (P 〉 0. 05 ). Serum TNF-α was significantly increased [ ( 8.59 ± 3.22 ) vs ( 5.29 ± 2.30 ) ng/L, P 〈 0.05 ] and serum IL-10 was significantly decreased [ ( 4.90 ± 1.32 ) ng/L vs ( 7.30 ± 2.14 ) ng/L,P 〈 0.05 ] in CHF group compared with that in PS group; After TU therapy,serum TNF-α level decreased markedly (5.97 ± 1.81 vs 8.59 ±3.22 ng/L, P 〈 0.05 ) and that of IL-10 increased markedly ( 6.55 ± 2.11 vs 4.90 ±1.32 ng/L, P 〈 0.05 ) . No significant change was found of sermn TNF-α and IL-10 levels in CHF + TU group according to those in PS group ( P 〉 0.05 ). Conclusion Low dose testosterone induces a reduction in TNF-α and an increase in IL-10 in male rats model of postinfarction CHF.
