小鼠呼吸道合胞病毒感染肺TLR4表达与气道高反应性 被引量:4

摘要 目的:研究小鼠呼吸道合胞病毒(RSV)感染对肺TLR4表达与气道高反应性的影响。方法:生后6周16只清洁级BALB/c小鼠被同等分成RSV感染组和对照组,感染后1周两组经吸入相同递增浓度乙酰甲胆碱检测气道高反应性,使用RT-PCR方法测定肺组织TLR4mRNA表达,并对二者的相关性进行了分析。结果:在吸入乙酰甲胆碱浓度为3.2g/L时两组气道阻力差异有显著性(P<0.01)。TLR4/β-actin的值分别为对照组0.239±0.055,RSV感染组0.361±0.102,两组间有显著性差异(P<0.05);TLR4 mRNA表达与吸入3.2g/L乙酰甲胆碱时的气道反应性呈正相关,相关系数为0.721(P<0.05)。结论:小鼠RSV感染后肺TLR4表达增强并呈气道高反应性,二者之间有显著正相关。 Objective: To study the lung tissue TLR4 expression and airway hyperresponsiveness(AHR) in the mice infected by respiratory syncytial virus(RSV). Methods: 16 clean, 6 weeks old BALB/c mice were divided into two groups equally(RSV group and control). One week after RSV infection, AHR was detected by inhaling the increased concentration of methacholine, TLR4 mRNA expression was measured by RT-PCR methods, and the correlation between AHR and TLR4 expression was also analyzed. Results:An important difference was detected in two groups' airway resistance when the inhaled concentration of methacholine was 3.2g/L(P 〈 0.01). The TLR4/β-actin was 0.361 ± 0.102 and 0.239 ± 0.055 respectively in RSV group and control group and there was a statistically significance(P〈 0.05). The expression of TLR4 mRNA was positively correlated with the airway hyperresponsiveness when the inhaled methacholine concentration reached 3.2g/L(r=0.721 ,P 〈 0.05). Conclusion:The lung tissue expression of TLR4 mRNA and AHR were increased in the mice infected by RSV and they were positively correlated.
