摘要
目的通过正交设计优化自然杀伤(NK)细胞最佳杀瘤条件。方法补体裂解法分离NK细胞,四甲基偶氮唑蓝(MTT)法测定NK细胞杀瘤活性,正交设计选择NK细胞最佳杀瘤条件。结果在葡萄球菌肠毒素B(SEB)浓度为10-7kg/L,SEB作用NK细胞时间为24 h,NK细胞与肿瘤细胞之比为20∶1条件下,NK细胞杀瘤活性最强。结论NK细胞最佳杀瘤条件组合为SEB 10-7kg/L,作用时间24 h,效靶比20∶1。
Objective To select the best combination of antineoplastic effect of NK cells by orthogonal design. Methods NK cells were .separated by complement lysis method, then monotetrazolium (MTT) was used to determine the antineoplastic effect of NK cells. The optimal combination was obtained by orthogonal design. Results On condition that SEB concentration was 10^-7kg/L,stimulating time was 12 h,and the ratio of NK cells and tumor cells was 20:1, the NK cell antineoplastic activity was the highest. Conclusion The best combination of antineoplastic effect of NK cells is followecl:SEB 10^-7kg/L,stimulating time 12 h,and the ratio of effect cells and target cells is 20: 1.
