MTT比色分析法在贴壁生长细胞药物敏感性测定中的应用

THE APPLICATION OF MTT COLORIMETRIC ASSAY TO MEASURE THE MEDICINAL SENSITIVITY FOR ADHESIVE CELLULAR GROWTH AND PROLIFERATION

用ＭＴＴ法（噻唑蓝比色分析法）测定了几种小檗胺类化合物对三种贴壁生长细胞（ＨｅＬａ、Ｂｏｗｅｓ、ＭＤＢＫ）生长增殖的影响．结果显示：ＭＴＴ分析法比传统的活细胞计数法、单细胞克隆法及３Ｈ－ＴｄＲ掺入法测定样品多、省时、省试剂、精确度高、重复性好及没有同位素污染等优点，是肿瘤药物研究中较理想的检测方法．

We have used MTT colorimetric assay to examine the effects of berbamine(B0)and its derivatives (B2,B4,EBB) on proliferation of three adhesive cell lines:HeLa,Bowes and MDBK cell.The results showed that the MTT colorimetric assay can examine a large number of samples in one time.MTT assay method was compared with the traditional assay methods, such as living cells counting,colong forming and 3H-TdR incorporation,the advantage of the former is its simplicity,rapidity, high degree of precision, reproducibility and no radioisotope pollution.Therefore the MTT colorimetric assay is a satisfying and prospective method on the study of anti-tumour drugs.
