南极冰藻Chlamydomonas L4的RPS5基因的获得及其在分类中的作用

摘要 通过对南极冰藻L4(Chlamydomonnas L4)cDNA文库的筛选,克隆出一个具有5’和3’非编码区的40S核糖体蛋白S5(RPS5)全长基因序列。通过与GenBank中日本凋毛藻、拟南芥、果蝇、斑马鱼、非洲爪蟾、人等物种的同源基因或ESTs序列比对,得到了根据RPS5基因的序列相似性绘制的系统分类树,其符合传统的物种分类系统。证明了通过持家基因的一种——RPS5基因的序列相似性进行分子分类学研究具有可行性。 A full-length 40S ribosomal protein S5(RPS5) gene sequence was cloned from Antarctic ice microalgae Chlamydomonas L4. Having compared with the homologous gene or ESTs sequence of Griffithsia japonica, Arabidopsis thaliana , Drosophila melanogaster , Danio rerio , Xenopus laevis and Homo sapiens in the GenBank, the taxonomic system tree based on the comparability of RPS5 genes was protracted, and the system tree is consistent with the traditional species taxonomic system, which indicates that the molecular taxonomy study based on the sequence comparability of RPS5 genes is feasible.
