Using Response Surface Methodology to Optimize the Separation of L-Ascorbic Acid and D-Isoascorbic Acid by Capillary Electrophoresis

摘要 采用三因素三水平的Box-Behnken设计,利用响应曲面法对影响毛细管电泳分离维生素C及异维生素C的主要影响因素(缓冲溶液的pH、浓度和分离电压)进行研究,以两种维生素C的分离度为响应函数,建立了相应的数学模型,得到了它们的最佳分离条件为:50 mM的三羟基甲基氨基甲烷-盐酸电泳缓冲液,pH=7.5,30 kV电压。 Response Surface Methodology ( RSM ) based on a three - level, three variable Box - Behnken design ( BBD ) was used to optimize the separation of L- ascorbic acid( L- AA) and D -isdascorbic acid( D -IAA) by capillary electrophoresis,with respect to important variables, namely, buffer pH values, buffer concentrations and operating voltages. Response surface methodology was used to establish the mathematical models using the resolution between two Vitamins as the responses. The optimum conditions were 50 mM Tris - HCI buffer and pH7.5 as the background electrolyte,and an applied voltage of 25 kV.
