摘要
为了开发一种用于治疗肿瘤的局部定位缓释给药系统,本研究在室温条件制备了生物可降解的聚乳酸(PLLA)纤维,并且详细研究了制备过程中的处方工艺条件(聚合物分子量、聚合物浓度、注射速度、出口口径以及固化液的种类)对聚乳酸纤维的成形特性(纤维直径、纤维的表面与内部结构以及纤维的结晶度)的影响。此外,还在聚乳酸纤维上上载了一种脂溶性模型药物(尼莫地平),并对载药纤维的体外释放性质进行了研究。结果表明,这类载药纤维可以成为一种有开发前景的局部定位给药缓释系统。
In order to explore a kind of local drug delivery sustained system to treat cancer, in this paper, a drug loaded PLLA biodegradable fiber was fabricated at room temperature. The influences of formulation and fabrication condition, such as polymer molecule weight, polymer concentration, pushing velrateity, needle diameter and nonsolvent, on the fiber's formation characteristic (fiber diameter, surface and inner structure and crystallinity) were intensively investigated. A hydrophobic model drug, nimodipine, was successfully loaded onto this fiber. Meanwhile, the drug release characteristics of the fiber in vitro were also studied. In conclusion, this drug loaded biodegradable fiber could be a promising local drug delivery sustained system.
