摘要
目的观察托吡酯对红藻氨酸(KA)诱导颞叶癫痫大鼠海马突触重建标记物神经细胞粘附分子(NCAM)表达的影响,进一步探讨托吡酯的抗痫作用机制。方法采用免疫组织化学染色观察KA诱导癫痫大鼠及托吡酯给药大鼠海马神经细胞粘附分子表达水平,并对两组NCAM表达进行比较。结果KA组NCAM表达水平各时间点平均NCAM阳性密度OD率均明显高于N S组和KA+TPM组(P<0.01)。结论本研究提示托吡酯可能通过抑制突触重建的形成,减少痫性放电,从而控制癫痫发作。
Objective To investigate the antiepileptic mechanism of topiramate(TPM) by observing the effect of topiramate on neural cell adhesion molecule in the hippocampus of temporal lobe epilepsy rats induced by kainic acid(KA). Methods The neural cell adhesion molecule was observed by immunohistochemical staining in the hippocampus of temporal lobe epilepsy rats induced by kainic acid and rats taking topiramate respectively. Results Compared with the control andTPM+KA groups,the average values of the ODin group KA were signifi cantly higher(P〈0.01). Conclusion Topiramate could perhaps control the seizures by reducing synaptic reorga nization and the epileptic discharge.
