人脑皮层神经元和NB4细胞对亚砷酸的耐受性差异 被引量:1

摘要 亚砷酸治疗急性早幼粒细胞白血病有效,但是因其难于透过血脑屏障而限制了防治中枢神经系统白血病的应用,在发明一种无创方法帮助其有效透过血脑屏障之前有必要明确其在中枢的安全有效的治疗浓度范围.应用激光共聚焦、琼脂糖凝胶电泳和蛋白免疫印迹技术,观察了砷干预前后人脑皮层神经元和白血病NB4细胞凋亡密切相关的分子标记物——胞质钙浓度([Ca2+]i)、DNA凋亡梯带和蛋白激酶C(PKC)活性的动态变化,同时应用酶免疫法(NSE-EIA)观察了砷干预前后神经元损伤的特异性标记物——神经特异性烯醇化酶(NSE)的变化.结果显示,1.0μmol/L的As2O3有效诱导NB4细胞分化和凋亡,而对皮层神经元没有明显影响.
