Mass spectrometry analysis of Jo-1 antigen

目的制备纯化的Jo-1抗原,进一步研究Jo-1/抗Jo-1系统的本质。方法从兔胸腺中提取粗制Jo-1抗原后经亲和层析纯化,采用液相色谱-电喷雾离子阱质谱分析肽谱和各肽段的氨基酸序列,经数据库检索鉴定Jo-1抗原的生化性质。结果用液相色谱-电喷雾离子阱质谱对Jo-1抗原进行分析,发现得分较高的有谷氨酰-tRNA合成酶和酪氨酰-tRNA合成酶。结论谷氨酰-tRNA合成酶和酪氨酰-tRNA合成酶有可能是Jo-1抗原的不同亚型。

Objective To purify Jo-1 antigen and to lucubrate the system of Jo-1 / anti - Jo-1 . Methods We extracted extractable nuclear antiger （ENA） from rabbit thymus, then purify Jo-1 antigen from ENA through affinity chromatography. Finally the purified Jo-1 antigen was analysed by liquid chromatography-electrospray ionization ion trap mass spectrometry/mass spectrometry （LC-ESI-IT-MS/MS）. Results By using LC-ESI-IT-MS, we found two proteins： glutamyl-tRNA synthetase and Tyrosyl-tRNA synthetase. Conclusion The two proteins may be isoforms of Jo-1 antigen.
