摘要
目的观察普罗布考对高脂兔脂肪肝形成的影响,并探讨其可能机制。方法16只新西兰大白兔给予高脂饲养8周后,随机分为2组:(1)淀粉组(n=8):饲以高脂饲料及1%淀粉;(2)普罗布考组(n=8):在饲以高脂饮食基础上给予1%普罗布考;另选8只兔为正常饮食组(n=8),给予普通饮食。14周末结束实验。测定血清脂质、超氧化物歧化酶(SOD)、丙二醛(MDA)和转氨酶水平,测定肝脏的脂质含量并进行病理学检测。结果(1)高脂饮食8周后总胆固醇、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇的含量明显升高,普罗布考可明显降低上述指标,均为P<0.01;(2)普罗布考对SOD活性无明显影响,但与淀粉组比较,普罗布考组MDA的含量[(9.2±3.3)μg/ml vs(12.6±4.4)μg/ml,P<0.01]明显降低;(3)普罗布考组血清转氨酶水平[ALT(45.3±12.9)U/L,AST(49.5±13.0)U/L]较淀粉组[ALT(67.20±14.3)U/L,AST(81.6±21.8)U/L]明显降低,均为P<0.01;(4)普罗布考干预6周后可明显降低肝脏中胆固醇和甘油三酯的含量,肝脏脂肪病变明显减轻。结论普罗布考具有降脂、抗氧化和护肝作用,能抑制高脂饮食所致的脂肪肝形成。
Objective To study the effect of probucol on the formation of fatty liver in high - cholesterol fed rabbits. Methods Sixteen male New Zealand white rabbits were fed with a diet composed of 1% cholesterol, 7.5% protein, and 8% lard for 8 weeks, and then were randomly divided into two groups: ( 1 ) starch group, maintained cholesterol diet plus 1% starch for 6 weeks; (2) probucol group, the same cholesterol diet supplemented with 1% probucol for 6 weeks. And control group was fed with normal diet for 14 weeks. We detected the changes in serum lipid profile, total activity of superoxide dismutase (SOD), maleic dialdehyde (MDA), alanine transaminase (ALT), aspartate transaminase (AST) concentrations, and the histopathological changes of the liver. Results Compared with control group, rabbits fed with high - cholesterol diet showed higher levels of serum total cholesterol (TC), low - density lipoprotein cholesterol (LDL- C) and HDL- C, all of which were significantly reduced by probucol. Probucol had no effect on the total activity of SOD, but MDA concentration was significiantly reduced as compared with starch group, ( all P 〈 0.05). The AST and ALT levels were dropped significantly at the P 〈 0.05 levels after six weeks' treatment with probucol. The fatty degeneration of liver was ameliorated in probucol group as compared with the starch group ( P 〈 0. 01 ). Conclusions Probucol has lipid - lowering and antioxidant properties and is significantly effective in protecting the liver function. Probucol can inhibit the formation of fatty liver in cholesterol - fed rabbits.
