摘要
目的:建立一种用气相色谱/质谱联用(GC-MS)技术测定绿康银杏叶片中α-亚麻酸的含量,以期完善其质量标准。方法:先将样品中紫苏籽油进行甲酯化后测定各脂肪酸的组成,并对总离子流图中各色谱峰相应的质谱图经人工解析图谱及计算机检索,确定其化学结构。色谱柱为FFAP30m×25mm×0.25μm;柱前压为80kPa;柱温为100~220℃;进样口温度为250℃。结果:共鉴定出4种脂肪酸,分别为棕榈酸、油酸、亚油酸、α-亚麻酸,相对百分含量分别为7.094%,21.032%,17.588%,54.285%。并且α-亚麻酸在0.2012~2.012g·L^-1范围内线性关系良好,r=0.9992,平均回收率为99.01%,RSD为1.35%;成品中的平均含量为1.111mg/片。结论:此种测定方法稳定性、重现性好,并且紫苏籽油中α-亚麻酸的相对百分含量最高,可以作为绿康银杏叶片的一项定量质控指标。
OBJECTIVE To develop a method to determine the α-linolenic acid in Lukang tablets Gingko lear tablets by gas chromatography-mass spectrometry (GC-MS) and to improve the quality standard. METHODS Fatty acids in the tablets were determined by GC-MS after methylesteration, and its total ions mass spectrogram was analyzed manually and retrieved by computer to determine its chemical structure. The column was packed with FFAP 30 m×25 mm×0. 25μm; anterior express was 80 kPa;column temperature was 100 - 220 ℃ ; injector temperature: 250℃. RESULTS 4 fatty acids were found in it, they were palmitic acid, oleic acid, linoleic acid, α-linoleic acid, and their relative contents were 7. 094%, 21. 032%, 17. 588%, 54. 285% respectively. The calibration curve of α- linoleic acid was linear within concentration range of 0. 201 2 - 2. 012 g·L^-1, r= 0. 999 2;The average recovery was 99.01%. RSD was 1.35%. The average content of α- linoleic acid in end-product was 1.1108 mg per-tablet. CONCLUSION This detective method is stable and reliable, the content of α-linoleic acid in Linseed oil is the highest and can be treated as a quantitative control marker.
