摘要
将香菇子实体干粉经热水浸提、乙醇沉淀,得到香菇多糖粗品。香菇多糖粗品经Sevage法去除游离蛋白,H2O2脱色,用DEAE-纤维素柱层析,经蒸馏水、0.05mol/LNaCl、0.1mol/LNaCl、0.2mol/LNaCl、0.3mol/LNaCl和0.4mol/LNaCl洗脱,得到6个香菇多糖级分A~F;然后用SephadexG-200柱进一步纯化香菇多糖B级分,得香菇多糖亚级分B-1和B-2;再研究其中的B-1。采用SephdexG-200柱层析和聚丙烯酰胺凝胶电泳法,证实该多糖组分为均一多糖。
Crude lentinan was obtained from fruitbody of lentinus edodes using following methods: extracting polysaceharide with hot water, precipitating with ethyl alcohol. Free proteins and pigment were removed by Sevage method and H202. Six lentinan fractions were got by DEAE-Cellulose column with eluent of water, 0.05 mol/L, 0.1 mol/L, 0.2 mol/L, 0.3 mol/L and 0.4 mol/L NaCl respectively. Letinan-B was further purified on Sephadex G-200 column, two subfraetions of letinan (B-1, B-2)were obtained. And letinan B-1 would be studied, too.Sephadex G-200 column chromatography and polyacrylamide gel eleetrophoresis illustrated that the fraction B-1 was homogenous polysaeeharide.
