摘要
通过碳源、氮源及主要营养与环境条件等对海洋红酵母产菌量的影响试验,以及蔗糖、蛋白胨、酵母膏三因素三水平的正交试验,获得了海洋红酵母的最佳发酵条件:蔗糖1.5%,蛋白胨1.0%,酵母膏0.5%,磷酸二氢钾0.15%,硫酸镁0.01%,氯化钠1.0%,初始pH5.0,接种量8%,500mL三角瓶中培养基装量为80mL,发酵温度为28℃,摇床200r/min。在此优化的培养条件下,海洋红酵母的产菌量可达9.7亿个/mL,比优化前发酵培养基产菌量(5.6亿个/mL)提高了73.2%,比单因素试验结果(8.1亿个/mL)提高了19.8%。
Through the carbon source, the nitrogen source and the main nutrition and the environmental condition and so on produce the fungus quantity to the Rhodotorula benthica the influence experiment, as well as the sucrose, the protein peptone, the yeast paste three factors three levels orthogonal experiments, we have obtained the Rhodotorula's best fermentation condition: sucrose 1.5 %, protein peptone 1.0 %, yeast paste 0.5 %, potassium dihydrogen phosphate 0.15 %, magnesium sulfate 0.01%, sodium chloride 1.0 %, initial pH 5.0, inoculating quantity 8 %, 80 mL medium in the 500 mL triangle bottle, temperature 28℃, 200 revolutions/minute. Under this optimized condition, the number of Rhodotorula has reached 970 million / mL, which enhanced 73.2 % to the initial condition (560 million/mL) and improved 19.8 % to the result of the test of the single factor (810 million/mL).
