酸性红-CTMAB-TX体系与DNA作用的共振光散射光谱的研究及分析应用 被引量:1

Resonance Light Scattering Spectrophtometic Determination of Trace DNA with the System of Fuchsine Acid-CTMAB-TX and its Analytical Application

摘要 研究了脱氧核糖核酸(DNA)与阴离子染料酸性红(FA)及阳离子表面活性剂溴化十六烷基三甲铵(CTMAB)和非离子型表面活性剂TritonX-10(0TX)作用的共振光散射光谱的特征.考察了各种影响因素,在优化条件下确立了共振光散射强度与鱼精DNA和小牛胸腺DNA以及变性后的鱼精DNA和小牛胸腺DNA浓度之间的关系,相应的线性范围分别为0.026-7.00mg/L;0.025-7.00mg/L;0.032-6.00mg/L;0.031-8.00mg/L,相关系数为0.9991;0.9996;0.9981;0.9996,检出限为7.0μg/L;6.0μg/L;11.0μg/L;12.5μg/L,基于共振光散射的增强,建立了一种测定DNA的新方法.图4,表3,参7. Resonance light-scattering (RLS) spectra characteristics of interaction between DNA and system of Fuchsine acid - CTMAB - TX was studied. The effects of interference substances and optimization of procedure were investigated and a new method was established for the determination of the fsDNA,ctDNA,denaturation fsDNA,and denaturation ctDNA at nanogram level based on RLS measurement. The linear range the determination are 0.026-7.00 rag/L; 0.025-7.00 mg/L; 0.032-6.00 mg/L; 0.031-8.00 mg/L and the correlation coefficient are 0.999 1 ;0.999 6;0.998 1 ; 0.999 6. The detection limit were 7.0 μg/L;6.0 μg/L; 11.0 μg/L; 12.5 μg/L. The method was applied to determination of DNA in mixed samples with satisfactory result. 4figs., 3tabs., 7refs.
