摘要
目的研究抑胶素对大鼠脑胶质瘤血管内皮生长因子(VEGF)的影响。方法采用脑立体定向术,在Wistar大鼠脑尾状核接种C6细胞,应用免疫组织化学方法检测应用抑胶素前后对鼠脑胶质瘤血管内皮生长因子的影响。结果经抑胶素治疗后可见肿瘤内血管内皮生长因子阳性细胞密度下降,随浓度增加阳性率呈下降趋势。实验组(抑胶素浓度32μg/kg)较对照组表达降低(P<0.05);实验组(抑胶素浓度64μg/kg)与对照组比较有显著性差异(P<0.01);而实验组、(抑胶素浓度分别为8μg/kg及16μg/kg)与对照组相比较,无统计学意义;而顺铂组瘤体内VEGF与实验组比较无显著性差异(P>0.05)。结论抑胶素对肿瘤细胞的抑制作用可能通过血管内皮生长因子而起作用,但这种抑制作用与抑胶素本身的浓度有关,高浓度抑胶素抑制作用强。
Objective To study the effect of antigliomatin on the expression of vascular endothelial growth factor (VEGF). Methods C6 glioma cells were injected into the caudate nucleus of rats stereotaxically. Treat the rats with Antigliomatin for 7d. The VEGF expression was detected by immunohistochemical methods before and after the drug treatment. Results Compared with the control group,the number of VEGF-positive cells was decreased,which was inversely correlated with the drug concentration. In the third experimental group(drug concentration: 32μg/kg),the positive VEGF cell number was lower than that in control group (P〈0.05). In the forth experimental group (drug concentration:64μg/kg),the positive VEGF cell number was much less (P〈0.01). There are no significance in VEGF positive cell numbers between the experimental groups (drug concentrations:8μg/kg, 16μg/kg respectively) and the control group. In the treatment group with cisplatin,the VEGF-positive cell number was decreased,just like that in the forth experimental group(P〉0.05). Conclusion It suggests that the inhibitive effect of antigliomatin on the growth of rat brain glioma is probably via down-regulating the VEGF expression. This kind of regulation is positively correlated with the drug concentration.
