摘要
研制了以Eu^3+为标记物的时间分辨荧光免疫分析仪。为使该分析仪灵敏性高、稳定性好,主要解决了2个关键性的问题:1)设计了理想的激发光系统,既能使光束最大效率地激发样品,又能保证杂散光对样品荧光的影响降到最小;2)设计了高效滤光系统,有效地降低了光谱噪声对光电倍增管的影响。同时,该分析仪利用稀土离子荧光寿命长、Stokes位移大的特点,采用了时间分辨技术,消除了散射光、短寿命杂质荧光的影响。经检验,该分析仪的灵敏度可达10^-12mol/L(当用Eu^3+为示踪标记物时),检测线性范围10^-12~10^-18mol/L,相关系数r=99.9%(P=0.0001),检测稳定性CV≤2.5%,是一种性能先进的用于超微量物质定量检测的临床检验仪器。
In the device,a pulsed Xenon lamp of flash frequency 1 kHz was adopted as exciting light,and the peculiarities of long lifetime and big Stokes about lanthanide trivalence ion and its chelates were utilized for the application of time-resolved and spectra-resolved technique. In order to make the system has high detecting sensitivity and nice measuring precision,two key technical problems in this device for TRFIA have been solved successfully. One is to design optimum exciting optical system which will make the light beam most effectively excite the sample fluorescence, and at the same time its stray light and mixed diffusion light minimally influence the sample fluorescence. The other is to have a perfect spectrum filter which makes the photomultiplier tube receives minimum spectrum noise. Thus the influence of scattering light and short-lifetime fluorescence was removed. By testing,the sensitivity is 10^-12 mol/L (when Eu^3+ was used for marking substance),examination repeat is 45%, examination linearity is from 10^-8 mol/L to 10^-12 mol/L,correlation coefficient is 99.9 % (p = 0. 0001 ). In a word,the instrument is advanced for ultrasensitive detection of antigen and antibody,and many other aspects.
