摘要
为证实化疗中血小板因子4(PF4)对正常骨髓细胞的保护作用,用体外培养人骨髓细胞,以MTT法测定细胞的存活率,观察PF4预处理后,骨髓细胞对化疗药物敏感性的变化。经PF4(5μg/ml)预处理48小时后,骨髓细胞对柔红霉素、阿霉素、鬼臼乙叉甙、氨甲喋呤和长春新碱的敏感性降低,药物的半数抑制浓度(IC50)增加(P<0.05),其作用与PF4的浓度及预处理的时间有关。结果提示,PF4能减轻化疗药物对正常人骨髓细胞的细胞毒性。
Objective:Toverifytheprotectiveefectofplateletfactor4(PF4)onnormalbonemar-rowcels.Method:AfterpreincubationofhumanbonemarrowcelswithPF4(5μg/ml)for48hoursinliquidculture,adrugwasaddedinaserialdilutionmannertogiveafinalvolumeof200μl/wel.Controlwelscontainedmediumwithoutdrug.ThecelsurvivalratesweremeasuredbyMTTassay.50%in-hibitoryconcentration(IC50)foraparticulardrugwasdefinedastheconcentrationproducing50%de-creaseincelgrowth.Result:PreincubationwithPF4causedanincreaseintheviabilityofcelsaftertreat-mentwithindividualdrugsinvariousconcentrationsandtheIC50ofeachdrugwasalsoincreased.Theef-fectswererelatedtothepreincubationtimeandtheconcentrationofPF4.Conclusion:PF4canprotecthumanbonemarrowcelsfromthecytotoxicityofchemotherapeuticdrugs,andapossibleuseofPF4asaprotectivedrugforthetreatmentofcancerwassuggested.
