摘要 目的探讨缺氧诱导因子HIF-1α、增殖细胞核抗原(PCNA)、Bcl-2蛋白在乳腺癌组织中的表达及相互之间的关系。方法用免疫组织化学SABC法检测60例手术切除后的乳腺癌石蜡标本组织中HIF-1α、PCNA、Bcl-2蛋白的表达情况,分析它们与患者年龄、月经状况、肿瘤大小、TNM分期、组织学分级、腋淋巴结转移情况、雌激素受体(ER)和孕激素受体(PR)水平等临床病理指标之间的关系,并探讨HIF-1α与PCNA和Bcl-2表达水平间的关系。结果HIF-1α阳性表达率为55%,其阳性表达与淋巴结转移、组织学分级成正相关;PCNA阳性表达率为76.67%,其阳性表达与肿瘤大小、组织学分级成正相关,与雌激素受体表达成负相关;Bcl-2阳性表达率为61.67%,其阳性表达与淋巴结转移、组织学分级成负相关,与雌激素受体表达成正相关。HIF-1α的表达与PCNA的表达无相关性,与Bcl-2的表达成负相关。结论HIF-1α、PCNA、Bcl-2的阳性表达率可作为判断乳腺癌转移与预后的重要指标。 Objective To investigate the expression of HIF -1α,PCNA and Bcl - 2 in breast cancer and their relationship. Methods The expression of HIF- 1α, PCNA and Bcl - 2 in paraffin embedment specimens of 60 breast cancers were detected by immunobistochemical SABC method respectively, and the relationship was analyzed between the expression and some clinical - pathological factors such as the age of onset, menstrual status, tumor size, clinical TNM stage, histological grade, axillary lymph node metastasis, levels of estrogen receptor (ER) and progesterone receptor (PR). And the relationship between HIF-1α, PCNA and Bcl- 2 was studied. Results The positive rate of HIF- 1α was 55% ,which expression was positively correlated with lymph node matastasis and histological grade ( P 〈 0.05). The positive rate of PCNA was 76.67%, which expression was positively correlated with tumor size and histological grade ( P 〈 0.05), and was negatively correlated with estrogen reccpter( ER). The possitive rate of Bcl - 2 was 61.67%, which expression was negatively correlated with 1)Tnph node matastasis and histological grade (P 〈 0.05), and was positively correlated with estrogen recepter(ER). To combine with these data, The expression of HIF- 1α was negatively conelated with Bcl- 2, while was not correlated with PCNA. Conclusion The expression of HIF -1α, PCNA and Bcl - 2 mav serve as important parameters for evaluating the malignant degree and prognosis of broast cancer.
