Apo H基因多态性与冠心病及血脂代谢的关系

RELATIONSHIP BETWEEN APOLIPOPROTEIN H GENE POLYMORPHISM WITH CORONARY HEART DISEASE AND LIPIDEMIA

摘要 目的探讨载脂蛋白H(ApoH)外显子3、8基因多态性与冠心病(CHD)及血脂代谢的关系。方法采用聚合酶链式反应结合限制性片段长度多态分析方法,分析了100例健康人及110例冠心病病人的ApoH外显子3、8基因型,并对血脂进行了测定。结果冠心病组ApoH外显子3GG、GA+AA基因型频率及G、A等位基因频率与对照组比较差异无显著性(X^2=1.56~2.15,P〉0.05)。冠心病组ApoH外显子8GC基因型频率及C等位基因频率显著增高(X^2=3.91、8.44,P〈0.05)。冠心病组ApoH外显子3GG、GA+AA基因型病人高密度脂蛋白胆固醇(HDL)显著高于对照组(t’=8.30,t=4.06,P〈0.05);冠心病组及对照组内基因型间的血脂水平比较无明显差异。冠心病组外显子8GG、GC基因型病人LDL水平也显著高于对照组(t’=7.97、8.55,P〈0.05);冠心病组GC基因型病人的三酰甘油(TG)水平显著高于GG型及对照组的各基因型(t’=4.02~4.71,P〈0.05)。结论ApoH外显子3基因多态性与冠心病及血脂代谢无相关性;ApoH外显子8GC基因型及C等位基因频率与冠心病发病有关,ApoH外显子8基因多态性与TG代谢有关。 Objective To study the relationship between the polymorphism of apolipoprotein H (Apo H) exon 3 and exon 8 with coronary heart disease (CHD) and lipidemla. Methods Polymerase chain reaction and technique of restriction fragment length polymorphism were used to detect the genotypes of Apo H exon 3 and exon 8 in 110 patients with CHD and 100 healthy subjects. Lipidemia was also detected. Results There was no difference between CHD group and control group in terms of the frequency of Apo H exon 3 GG, GA--AA genetype, and G and A allele frequency (X^2= 1.56-2. 15,P〉0.05); the frequency of GC genetype and C allele of Apo H exon 8 in CHD group was more higher than in the control group (X^2= = 3.91,8. 44 ;P〈 0.05). LDL of Apo H exon 3 in the CHD group was higher than in the control group (t'=8.30,t=4.06,P〈0.05). There was no difference of lipidemia of other genetype in CHD group and the control group. LDL of Apo H exon 8 in CHD group was more higher than that in control group (t'=7.97,8.55;P〈0.05). TG of Apo H exon 8 in CHD group was more higher than GG genetype and other genetype of control group (t'= 4.02 --4.71, P〈 0.05). Conclusion There was no relativity between Apo H exon 3 gene polymorphism and CHD and lipidemia, Apo H exon 8 GC genetype and C allele have something to do with CHD, and Apo H exon 8 gene polymorphism have something to do with TG.
