慢性粒细胞白血病端粒酶活性检测的临床意义 被引量:1

CLINICAL SIGNIFICANCE OF DETECTION OF TELOMERASE ACTIVITY IN CHRONIC MYELOID LEUKEMIA

摘要 目的探讨端粒酶活性检测对慢性粒细胞白血病(CML)的临床意义。方法选取20例慢性期和10例急性转化期(加速期或急变期)的CML病例,以良性血液病病人和正常人作对照,采用改良TRAP-银染法及亚利恩凝胶成像系统分析检测其骨髓标本内的端粒酶活性水平并随访。结果良性血液病端粒酶阴性或低水平表达;CML慢性期端粒酶活性中等增高(F=36.38,q=5.84,P<0.01),阳性率达35%(7/20);急性转化期时端粒酶活性较慢性期显著升高(q=12.06,P<0.01),阳性率达80%(8/10)。端粒酶活性水平与CML预后相关,活性高者往往预后较差。结论端粒酶活性与CML的发生发展密切相关。端粒酶活性检测可以作为CML诊断、病程监测及预后判断的一个有用的生物学指标。 Objective To assess the clinical significance of telomerase activity in chronic myeloid leukemia(CML). Methods Using modified TRAP silver staining assay and Yallen analytical software, the telomerase activity in bone marrow samples from 20 CML patients in chronic phase and 10 patients in acute transformation were measured. Patients with benign hematopathy and non-hematopathy with normal bone marrow figures served as controls. Results In benign hematopathy, telomerase expression was negative or low; in chronic phase CML, telomerase activity increased moderately~ in acute transformation, the activity increased significantly. High telomerase activity suggested poor prognosis. Conclusion Telomerase activity is closely related to the occurrence and development of chronic myelocytic leukemia. The detection of telomerase activity can be used as a biological marker for diagnosis, monitoring, and prediction of the disease.
