Survivin反义寡核苷酸诱导胃癌SGC7901细胞株凋亡的实验研究

摘要 目的 观察Survivin反义寡核苷酸(ASODN)对胃癌细胞株(SGC7901)增殖、凋亡的作用。方法 人工合成Survivin硫代ASODN,通过脂质体转染胃癌细胞株(SGC7901)后,用MTT法检测细胞毒作用;用荧光染色、流式细胞术检测细胞凋亡;用RT-PCR、免疫组化技术检测SurvivinmRNA及蛋白的表达。结果 (1)Survivin ASODN抑制胃癌SGC7901细胞增殖呈时间和剂量依赖性;(2)Survivin ASODN处理组SGC7901细胞24h后凋亡率为33.6%,正义寡核苷酸(SODN)组为10.7%,2组相比,有显著性差异(P〈0.05);(3)SurvivinASODN处理组SGC7901细胞Survivin蛋白及mRNA的表达水平显著下降。结论 Survivin ASODN能够抑制增殖、诱导凋亡,推测可能通过下调Survivin mRNA及蛋白表达而起作用。
