摘要
在培养基中添加不同浓度的卡那霉素和壮观霉素,观察抗生素对甜菜叶柄分化再生的影响。结果表明:一定浓度的卡那霉素和壮观霉素对甜菜叶柄的分化均具有抑制作用,当卡那霉素浓度为50mg/L时,叶柄分化率明显变化,当浓度达到100mg/L时,分化率为1.4%。当壮观霉素浓度为40mg/L和50mg/L时,分化率为5.7%和3.7%,当浓度为60mg/L时,基本无绿芽分化,统计学分析显示:壮观霉素的抑制作用要强于卡那霉素。结合实验结果讨论了这两种抗生素作为甜菜遗传转化研究的筛选标记的适宜浓度。
This paper studied the effect of the supplement of kanamycin and spectinomycin on regeneration of sugar beet petioles. The results indicated that regeneration of sugar beet petioles was inhibited by the two antibiotics with certain concentrations.When the concentration of kanamycin was 50 mg/L, the petiole shoot frequency changed obviously. Particularly,the frequency were only 1.4% with 100mg/L kanamycin. When the concentrations of spectinomycin were 40 mg/L and 50mg/L, the frequencies were only 5.7% and 3.7%. Few green petiole shoots were regenerated with 60mg/L spectinomycin. Statistical analysis indicated the toxicity of spectinomycin was more serious than that of kanamycin. According to data from the research, we discuss the appropriate concentrations of the two antibiotics as selecting markers used in production of transgenic sugar beets.
