p21基因对NIH3T3细胞生长的抑制作用

摘要 为了证明p21是抑制细胞生长所必须的因子,我们用p21融合基因载体转染NIH3T3细胞,观察它的形态变化和生长周期,免疫印迹检测p21的表达和其下游蛋白PRb的磷酸化情况.结果显示NIH3T3细胞出现了衰老样变化,生长周期延长,生长速度缓慢;在转染了p21基因的NIH3T3细胞中PRb磷酸化程度降低.实验表明,过量表达的p21基因是通过降低PRb磷酸化水平抑制DNA的转录来抑制细胞生长的. In order to prove that p21 was essential gene to restrain the growth of cell, used p21 fusion gene vector to transfect N IH3T3 cell, observed the morphology changes and growth cycle, the expression of p21 and the varation of downriver protein phosphorylation were checked by immunoblotting. The result showed that NIH3T3 cells revealed premature Senescence,growth cycle prolonged and growth speeding slowed, prb' sphosphorylation degree in NIH3T3 cells infected by p21 genes falled, This test showed that excessive expression p21 genes inhibited the growth of cell through debasing PRb phosphorylation degree which leaded to inhibit transcription of DNA.
