温郁金对胃癌细胞的抑制作用及其对血管内皮生长因子表达的影响 被引量:33

摘要 目的:探讨温郁金对人胃癌细胞SGC-7901的抑制作用及其作用机理。方法:采用超临界二氧化碳萃取法提取温郁金成分;以5-Fu为阳性对照药物,采用MTT法,测定温郁金提取物对体外培养胃癌细胞SGC-7901增殖的影响;运用半定量逆转录多聚酶链反应(RT-PCR)方法检测温郁金提取物对胃癌细胞血管内皮生长因子(VEGF)mRNA表达的影响。结果:温郁金提取物对胃癌细胞的生长有显著的抑制作用(均P<0.01),且抑制率随药物浓度的升高而增高,存在明显的量效关系;100mg/L、200mg/L浓度的温郁金提取物对胃癌细胞生长的抑制率分别为51.59%、64.44%,在这个浓度水平,其抑制肿瘤细胞生长作用与阳性对照药物5-Fu效果相当。温郁金提取物能下调VEGF121和VEGF165 mRNA表达。结论:温郁金提取物对人胃癌细胞SGC-7901生长有抑制作用,其抑癌作用的机制可能与下调VEGF表达水平有关。 Objective :To study the inhibition and the mechanism of common turmeric (wenyujin) on human gastric carcinoma cell line SGC -7901 in vitro. Methods :The extract of connnon turmeric was obtained by supercritieal fluid extraction (SPE). With the 5 - FU as the positive control, tlle effects of the extract of common turmeric on proliferation of human gastric carcinoma cell line SGC-7901 in vitro by using MTT assay were studied. The effects of common turmeric on vascular endothelial growth factor (VEGF) mRNA expression in human gastric careinonla cell lille SGC -7901 were investigated by employing the semi - quantitative method of reverse transcription polymerase chain reaction ( RT - PCR). Results : Compared with the negative control, extract of common turmeric can significantly inhibit the growth of human gastric carcinoma cell line SGC -7901 in vitro with certain concentration( all P〈0.01 ) ,and the inhibitory rates for growth of SGC -7901 were positively con'elated with the concentration. In 100 mg/L and 200 mg/L level, the inhibitory rate, was 51.59% and 64.44%, respectively, there were no different between the extract of common turmeric and 5 - FU in tlleir inhibition on SCC-7901(P〉0.05). Incubated with Concentration at 100 mg/L of common turmeric, the expression VEGF121 mRNA and VEGFI65 mRNA in SGC-7901 cell deceased gradually( all P〈0.01 ). Conclusion : The eonlmon turmeric could inhibit the growth of human gastric carcinoma cell line SGC - 7901 in vitro, the decrease of VEGF expressiou might be the primary mechanism.
