家族性胆固醇结石病人SCP_2基因表达的研究

Expression of sterol carrier protein 2 gene in patients with hereditary cholesterol gallstone

目的研究具有家族性胆囊胆固醇结石病人、非家族性胆囊胆固醇结石病人和非结石病人肝组织 SCP_2基因的表达情况。方法应用逆转录-聚合酶链反庸(RT-PCR)技术,研究了28例具有家族遗传性胆囊胆固醇结石病人、30例非家族性胆囊胆固醇结石病人和32例非胆固醇结石病人肝组织 SCP_2 mRNA 的表达水平;同时应用 Western blotting 法分别检测三组病人肝组织中 SCP_2蛋白含量变化;并采用生化技术行胆汁脂质分析,计算成石指数。结果家族遗传性和非家族遗传性胆固醇结石组 SCP_2 mRNA 表达水平较非胆固醇结石升高,差异有显著性;家族遗传性胆固醇结石SCP_2 mRNA 表达水平较非家族性胆固醇结石升高,差异有显著性:SCP_2蛋白水平的变化与 SCP_2mRNA 变化一致,差异有显著性。结论胆囊胆固醇结石病人肝组织 SCP_2mRNA 表达水平升高,SCP_2基因表达上调可能是胆囊胆固醇结石形成的重要原因;SCP_2基因表达水平上调则可能在遗传中起一定作用。

Objective To investigate the expression of sterol carrier protein 2 （SCP2） gene in patients with hereditary cholesterol, non-hereditary cholesterol or non-cholesterol gallstone. Methods The expression of SCP2 mRNA was determined with RT-PCR in 28 patients with hereditary cholesterol gallstone, 330 with non-hereditary cholesterol gallstone and 32 with non-cholesterol gallstone. Meanwhile, the level of liver SCP2 was determined by Western blotting in these patients. Results The expression level of SCP2 mRNA was significantly higher in the patients with cholesterol gallstone than in those with non-cholesterol gallstone. Moreover, it was markedly higher in the patients with hereditary cholesterol gallstone than in those with non-hereditary one. Conclusions SCP2 is over-expressed in the patients with cholesterol gallstone, which suggests that SCP2 might be an important cause of cholesterol gallstone. Meanwhile, the over-expression of SCP2 may be a hereditary factor during the formation of cholesterol gallstone.
