克山病病区粮喂养大鼠红细胞膜脂流动性的研究

A Study on Lipid Fluidity of Erythrocyte Membrane of Rats Fed Selenium Deficient Diet

本文观察了低硒的克山病病区粮和克山病病区粮补硒后喂养大鼠其红细胞膜脂流动性的变化。实验结果表明克山病病区粮喂养的大鼠红细胞膜脂流动性较正常对照降低,其原因可能与机体处于低硒状态下红细胞膜结合硒含量降低、红细胞膜胆固醇含量及脂质过氧化产物升高有关;克山病病区粮补硒后喂养大鼠其红细胞膜脂流动性可恢复至正常对照。

A comparative study on the change of lipid fluidity of erythrocyte membrane (EM) of rats fed Se-deficient diet (Se content: 0.024ppm) and Se-supplement diet (supplement Se 0.25ppm as Na2SeO3) respectively was carried out in order to study the effect of low Se status on the membrane lipid fluidity. The rats fed the diet (Se content: 0.l05ppm) from the Animal Center of our university were taken as the controls. Lipid fluidity of EM was measured by the fluorescence polarization method using diphenylhexatriene as a fluorescent probe. The results showed that lipid fluidity of EM of rats fed the Se-deficient diet was significantly lower than that of controls; the selenium contents of EM and plasma were reduced; the contents of cholesterol, sialic acid and lipid peroxide of EM were increased; the activity of the whole blood GSH-Px was decreased. These changes could be prevented or returned to the control level when the rats were fed with Se-supplement diet. Based on the results of our study, it is concluded that the decrease in lipid fluidity of EM of rats fed Se-deficient diet is probably related to the reduce of the membrane selenium content and the increase of cholesterol and lipid peroxide of the membrane.
