甲状腺转录因子-1(thyroidtranscriptionfactor-1,TTF-1)是调节SP-B基因表达的关键因子。最近研究发现了一种新的蛋白即TTF-1相关蛋白(TAP26),它不仅能与SP-B基因启动子近侧序列相互作用,而且还能与TTF-1形成蛋白复合物,促进SP-B基因的表达。采用染色质免疫沉淀分析证实TAP26是肺细胞中SP-B启动子转录复合物的一个组成部分,为SP-B基因表达的调控提供分子机制。
Thyroid transcription factor-1 (TTF-1) is a known key factor that controls the gene expression of surfactant protein-B (SP-B). Recently a novel transfactor, a 26 ku protein was identified and now designated as TTF-1-associated protein 26 (TAP26). It interacts with the SP-B promoter fragment, forms a protein complex with TTF-1 in vitro and is capable of enhancing human SPB promoter activity in vivo. Using the chromatin immunoprecipitation assay (CHIP), it was demonstrated that TAP26 was a component of the transcription complexes of the SP-B promoter. It is a newly found molecular mechanism for the regulation of SP-B expression.
