摘要
目的探讨环孢素A(Cyclosporine A,CsA)对阿霉素(Adriamycin,ADM)治疗肝癌的影响。方法培养HepG2肝癌细胞,分成对照、CsA、ADM、CsA+ADM组,分别处理24V~48h,采用MTT法检测细胞增殖、流式细胞术测定细胞周期及细胞凋亡。结果肝癌细胞增殖能力48h明显受到抑制,CsA在5~1000μg/L随浓度增加,凋亡率逐渐增加,超过该浓度时,则出现凋亡率减低。CsA、及ADM均使G2期显著延长;CsA对HepG-2肝癌细胞有促进细胞凋亡作用.与ADM有协同促调亡作用。结论环孢素A对肝癌增殖有抑制作用,使G2期阻滞,促凋亡。环孢素A联合阿霉素对肝癌治疗有协同作用。
Objective To investigate the influence of combined cyclosporine A with adriamycin on hepatocellular carcinoma. Methods HepG-2 cells were cultured, they were divided into 4 grops namely, control.CsA.ADM,CsA+ ADM group, acted on respectively for 24 to 48 hours. Cell growth was assessed with MTT, cell cycle and cell apoptosis were assessed by flow cytometry. Results The ability of growth was inhibited in 48 hour. CsA and ADM could increased cell ratio of G2 phase; the apoptosis ratio was increased when CsA was below 1000μg/L, and when they were beyond the value, the apoptosis ratio was decreased. CsA combined with adriamycin demonstrated synergistic role effect on apoptosis. Conclusion CsA could inhibit the growth of hepatocellular carcinoma, arrest G2 phase, increased apoptosis. CsA combined with adriamycin demonstrated synergistic role effect on hepatocellular carcinoma.
