中药鸦胆子总苦木内酯提取物中鸦胆苦醇的含量测定

Determination of the content of brusatol in general quassinoids of fruits of Brucea javanica(L.) Merr.

目的建立RP-HPLC法测定中药鸦胆子总苦木内酯提取物中鸦胆苦醇的含量。方法采用RP-HPLC法：色谱柱为C18柱（250mm×4．6mm，5,um），流动相为甲醇-水（体积比为41：59），检测波长为277nm，柱温34℃。结果鸦胆苦醇质量浓度在43．6-348．8mg·L^-1内与峰面积呈良好的线性关系，r=0．9999（n=5），平均回收率为100．7％，RSD为1．6％（n=9）。结论测定方法准确、可靠，为鸦胆子总苦木内酯提取物质量评价提供了可靠的依据。

Objective To establish a HPLC method for the determination of brusatol in general quassinoids of fruits of Brucea javanica （L.） Merr. Methods A C18 column（250 mm×4.6 mm, 5 μm）with a solvent system consisting of methanol-water （ V ： V = 41 ： 59） was used, and the detecting wavelength was set at 277 nm. The column temperature was 34 ℃. Results The calibration curve was linear in the range of 43.6- 348.8 mg.L^- 1 with the correlation of 0. 999 9 （ n = 5）. The average recovery of brusatol was 100.7 % with RSD of 1.6 % （ n = 9）. Conclusions The method is sensitive, accurate, rapid, and is suitable for the quality control of the general quassinoids of fruits of Brucea javanica （L.） Merr.
