摘要
目的研究MMP-3、MMP-8、在翼状胬肉发生发展中的作用及其相关性和临床意义。方法①10例翼状胬肉标本抽提总DNA并行RT-PCR扩增,探讨MMPs基因表达在翼状胬肉中的作用。②应用免疫组织化学S-P法检测50例翼状肉组织和10例正常结膜组织MMP-3、MMP-8的表达,并分析其与翼状胬肉临床病理参数间的关系。结果1.MMP-3在10例正常结膜组织中未见阳性表达,50例翼状胬肉组织中阳性~强阳性表达39例,阳性率78%,与正常结膜组织间差异具显著性(p<0.005)。MMP-3在翼状胬肉组织的进展期、静止期和复发性病变组织的表达,差异具显著性(p<0.05)。在不同职业患者的MMP-3表达有显著性差异,其中户外MMP-3的表达高于户内(p<0.05)。在不同年龄组、性别组MMP-3表达无显著性(p>0.05)。2.MMP-8在10例正常结膜上皮组织中阳性表达1例,而50例翼状胬肉组织中,阳性~强性性表达41例,阳性率82%,与正常结膜组织间差异具显著性(p<0.005)。在不同职业患者的MMP-8表达有显著性差异,其中户外MMP-8的表达高于户内(p<0.05)。在不同分期及复发性的病变组织中、在不同年龄组、性别组MMP-8表达差异均无显著性(p>0.05)。3.MMP-3与MMP-8的表达具有高度相关性(p<0.005)。结论MMP-3、MMP-8在翼状胬肉中表达的程度与其患者的职业类型有关,但与患者的性别、年龄无显著相关性。MMP-3的表达与其分期及复发相关,MMP-8的表达与其分期及复发不相关。MMP-3与MMP-8的表达具有高度相关性,它们之间可能有协同作用。MMP-3、MMP-8活性表达升高可能与翼状胬肉的发生发展有关。
Objective To study the role and relationship of MMP-3, MMP-8 in pterygia and to evaluate the clinical significance. Methods 1.10 pterygium specimens were collected and the total RNA were extracted and amplified the gene fragments to investigate the role of MMP played in pathogenesis of pterygium. 2. The expression of MMP-3 and MMP-8 was examined by immunohistochemical technique in 50 cases of pterygia and 10 cases of normal conjunctival tissues. The relationship between the expression and clinicalpathological parameters were also analyzed. Results 1. The positive rate of MMP-3 was 0% in 10 cases of normal tissues and 78% in 50 cases of pterygia, namely 39, the difference between them had markedly statistical significance (p〈0.005), the staining intensity of MMP in pterygia was significantly associated with the clinicopathologic stage and recurrence (p〈0.05). There was significantly different in the expression of MMP-3 among various occupations, higher outdoors (P〈0.05), but no distinct difference was found in other parameters. 2. The expression of MMP-8was much higher in pterygia than in normal tissues (p〈0.005), the positive rate was 82% and 10% respectively, it had no relevancy with the clinical stage and recurrence (p〉0.05). There was significantly different in the expression of MMP-8 among various occupations, higher outdoors (p〈0.05), but no distinct difference was found in other parameters. 3. There was significant correlation between the expression of MMP-3 and MMP-8 (p〈0.005). Conclusions The expression of MMP-3, MMP-8 is correlated with occupation, but no distinct difference with clinical parameters such as sex and age. The expression of MMP-3 in pterygia is significantly associated with the clinicopathologic stage and recurrence, but that of MMP-8 is not. there may beco-operated effects between MMP-3 and MMP-8. These enzymes may be responsible for the progression of pterygia.
