摘要
目的研究有丝分裂原激活蛋白激酶(MAPK)-p38在佐剂关节炎(AA)大鼠滑膜及肺组织中表达,探讨类风湿关节炎(RA)的发病机制。方法给大鼠右后足跖部皮下注射完全弗氏佐剂(CFA)复制RA的动物模型-AA。用原位杂交法检测p38mRNA在AA大鼠滑膜及肺组织中的表达。结果AA大鼠滑膜和肺组织p38mRNA阳性细胞的平均灰度值(0~255,深~浅)明显低干正常大鼠(P〈0.01)。AA大鼠滑膜的巨噬细胞,成纤维细胞和淋巴细胞的胞浆内均有p38mRNA的表达。肺组织中p38mRNA主要表达于巨噬细胞,单核细胞及浸润的淋巴细胞中。结论AA大鼠p38mRNA过表达与AA的滑膜及肺部病变有关,可能在RA的发病中有重要作用。
Objective To explore the pathogenesis of rbeumatoid arthritis (RA) by studying tbe expression of mitogen activated protein kinase ( MAPK)-p38 in the synovium and lung of adjuvant arthritis rats. Methods To build animal model of RA by injection of CFA. The expression of p38 mRNA was detected by in situ hybridization and analyzed by microimage analysis system. Results The average grey values (0 - 255, black - white) of p38mRNA positive cells in synovium and lung of the arthritis rats were much lower than those in the nonnal control rats (P 〈 0. 01 ) , p38 mRNA positive cells were the macrophage cells, fibroblast cells and lymphocytes in the synovium. In the lung tissue, p38 mRNA expressed in the mcroeytes, monocytes and tymphocytes. Conclusion The overexpression of p38 maybe related to the synovium and pulmonary affection of RA and may play an important role in the pathogenesis of RA.
