基质金属蛋白酶-9对大肠癌细胞肿瘤坏死因子受体1表达的影响

摘要 目的研究基质金属蛋白酶9(MMP-9)对大肠癌细胞肿瘤坏死因子受体1(TNFR1)表达的影响以及促进大肠癌侵袭和转移的可能途径.方法通过免疫荧光法研究TNFR1在大肠癌细胞SW1116中的表达;用MMP-9干预培养的大肠癌SW1116细胞,用流式细胞术检测MMP-9不同剂量和不同作用时间时TNFR1表达变化.结果①SW1116细胞表达TN-FR1;②MMP-9对SW1116细胞表面TNFR1表达有下调作用,但有剂量和时间依赖性.结论MMP-9下调大肠癌细胞表面TNFR1的表达,可能与大肠癌侵袭转移密切相关. AIM: To study the effects of matrix metalloproteinase-9 (MMP-9) on the expression of tumor necrosis factor receptor-1 (TNFR-1) and migratory and invasive potentials in human colon carcinoma cells. METHODS: Immunofluorescence staining was applied to examine the expression of TNFR-1 in SW1116 in human colon carcinoma cell lines. The expression of TNFR-1 was assayed by flow cytometry in human colon carcinoma SW1116 cells treated with MMP-9 at different concentrations and at different time periods of culture. RESULTS: TNFR-1 was expressed in SW1116 cells and MMP-9 down-regulated the expression of TNFR1 on cell surface in a dose- and time-dependent manner. CONCLUSION: MMP-9 decreases the expression of TNFR1 on the surface of human colon carcinoma cells, which may be a factor associated with invasive and metastasis potentials of colon carcinoma.
