雌激素受体α和βmRNA变异型在胎盘组织的表达及定位

Expression and Localization of Estrogen Receptor ERα and the Kinds of ERβ mRNA on Placenta

目的探讨人足月胎盘组织雌激素受体α、β(ERα、ERβ)的mRNA变异型种类及其在胎盘的组织细胞学定位。方法采用逆转录聚合酶链反应技术和免疫组织化学技术检测正常足月妊娠胎盘组织中ERα、ERβ表达及细胞学定位。结果人足月胎盘组织中有ERα、ERβmRNA的表达,胎盘组织中ERβmRNA的表达为ERβ5变异型;ERα表达定位于滋养层细胞滋养细胞核,ERβ表达定位于滋养层合体滋养细胞胞浆。结果ERβ5是胎盘组织ERβ的变异型种类,定位于胎盘合体滋养细胞,与胎盘合成和分泌雌激素有关。

Objective To investigate the kinds of ERa, ERfl mRNA and the localization of ERα, ERβ on normal human placenta. Methods The expression and localization of ERα,ERβ on normal human placenta were detected by reverse transcription-ploymerase chain reaction （RT-PCR） and immunhistochemistry （SP-assay）. Results There was mRNA expression of ERα and ERβ on human term placenta, and ERβ was identified with ERβ5. The ERα immunoreactivity on human term placenta was in villous cytotrophoblastic nucleus, and that of ERβ was in syncytiotrophoblastic cytoplasm. Conclusion The findings indicate that ERβ5 is the kind of ERβ on placenta. It is localizated in the cytoplasm of syncytiotrophoblast and is probably connected with the production of estrogen.
