摘要 目的:探讨结直肠癌(colorectal cancer,CRC)组织淋巴管生成情况与血管内皮生长因子-C(vascular endothelialgrowth factor-C,VEGF-C)和VEGF-D的关系。方法:应用淋巴管特异标志podoplanin检测96例结直肠癌及正常结直肠组织中淋巴管密度,以CD34标志血管作对比研究,并分别与ki-67进行双标免疫组化染色检测淋巴管和血管增殖活性;免疫组化SP法检测VEGF-C和VEGF-D的表达。结果:VEGF-C和VEGF-D表达与结直肠癌微淋巴管密度(microlymphatic den-sity,MLD)密切相关(P<0.05,P<0.01),尤以癌组织边缘区为甚(P<0.01),并且VEGF-C和VEGF-D高表达的结直肠癌组织中淋巴管内皮细胞ki-67指数也显著增加(P<0.01)。结论:结直肠癌组织存在新生淋巴管;VEGF-C和VEGF-D高表达可能诱导结直肠癌尤其癌周淋巴管生成。 Objective: To explore the relationship between lymphangiogenesis and expressions of vascular endothelial growth factor (VEGF)-C and VEGF-D in colorectal cancer (CRC). Methods: Specimens were obtained from 96 patients with CRC. Microlymphatic density (MLD) and microvessel density (MVD) in tumor and normal tissues were evaluated immunohistochemically with anti-human podoplanin and anti-CD34 antibody, respectively, ki-67/ podoplanin and ki-67/CD34 double immunostaining method were used to detect proliferating lymphatic vessels and blood vessels, respectively. The expressions of VEGF-C and VEGF-D were determined by SP immunohistochemical method. Results: VEGF-C and VEGF-D expressions were significantly correlated with MLD in CRCs (P〈0.05, P〈0. 01) ,especially in the margin region (P〈0.01). Futhermore, tumor lesions with higher VEGF-C and VEGFD expressions showed an increased ki-67 index in lymphatic endothelial cells (P〈0.01). Conclusion: There exist neonatal lymphatic vessels in CRC. VEGF-C and VEGF-D may induce lymphangiogenesis especially in the margin region of CRC.
