摘要
采用山羊不同免疫组织作为原料,以氯化钠-柠檬酸钠法与苯酚-稀盐法进行RNA提取的比较;分别确定两种方法对于不同免疫组织(肝脏、脾脏和淋巴)最大提取率分别为0.156%、0.150%、0.182%和0.142%、0.127%、0.164%。采用华特生公司蛋白质浓度检验试剂盒BCA法测定蛋白含量。定磷法测DNA含量,地衣酚显色法测RNA含量,样品用紫外分光光度计进行扫描,苯酚-稀盐法提取淋巴组织,RNA紫外吸收OD260:OD280=2.042077;氯化钠-柠檬酸钠法提取淋巴组织RNA紫外吸收OD260:OD280=2.1098。
Different goat immune tissues were used as raw materials , RNA was extracted by sodium chloride - citric sodium and phenol - the thin salt methods. The most extraction rate was 1.56‰ , 1.50‰ , 1.82‰ and 1. 42‰ , 1.27‰ , 1.64‰ respectively from different immune tissues( liver,spleen and lymph). The result showed that the phenol -thin salt method was better.
