摘要
经连续六代紫外幅射诱变和筛选,获得能连续六代稳定生产高果胶酶活性的突变体GBIB U7.研究了该突变体的果胶酶形成特性.实验结果表明,丰富的多糖,无机氮源及适量的阴离子去污剂培养基有利于突变体GBIB U7提高果胶酶活性.在液体摇瓶培养时,144小时左右果胶酶活性最高.以果胶为诱导物,该突变体的果胶酶诱导阈值为1.25×10(-6)M果胶,在乳糖存在的条件下,其诱导阈值降低.
The mutant strains, GBIB U7 with higher production of pectinases were obtained by screening of GBffi O7 on the solid media repeately after irradiation of the strains by ultraviolet ray more than six generation, the mutants of the sixth generation still have a stable ability of producing pectinases. The factors effecting of production of pectinases were investigated. Polysaccharose, inorganic nitrogen, and suitable detergent benefited the production of pectinases. The optimal time for pectinase production in liquid media was at about 144-th hour. The critical concentration of inducing pectinase production in the mutants was baout 1.25 X 10-6M of pectin, while in presense of lactose, the concentration was decreased.
