摘要
目的比较不同剂量的转化生长因子β1(TGF-β1)关节内注射对关节软骨全层缺损修复在细胞形态学、细胞基质化学方面的不同影响。方法将兔膝关节造成关节软骨全层缺损,分别注入1ng、10ng1、00ng TGF-β1,2、4、8周后行组织学、组织化学、免疫组化研究,分析比较修复组织中透明软骨细胞数和胶原含量的差异。结果各剂量组均有一定的修复效果,100ng组最佳,但出现了骨软骨赘和正常关节软骨的退变。结论实验各剂量TGF-β1都可以促进关节软骨全层缺损的修复,100ng较理想,最佳剂量和方法仍需探讨。
Objective To compare the effects of different doses of transforming growth factor-betal(TGF-β1 ) on articular cartilage defect(ACD) in terms of cytomorphology and cytostromatic chemistry. Methods ACDs were made in the knee joints of rabbits and lng, 10ng and 100ng of TGF-β1 were injected into them. Histological, histochemical and immunohisto chemical research was conducted two, four and eight weeks later to compare the number of true cartilage and the content of collagen. Results Different doses produced reparing effects of different degree. Best results were got in the group treated with 100ng, but degeration of normal cartilage and osteophyte occurred. Conclusion Different doses of TGF-β1 can produce different curatire effects with the lOOng as the best one in this experiment,but the optimal dosage and the treament still need to be further explored.
