瘦素对子宫内膜间质细胞分泌白细胞介素的影响

摘要 目的:观察瘦素对体外培养人子宫内膜间质细胞(ESC)分泌白细胞介素-6(IL-6)和白细胞介素-8(IL-8)的影响,探讨瘦素对女性生殖生理的调节作用。方法:采用ELISA方法测定加入不同浓度瘦素时ESC分泌IL-6和IL-8的含量。结果:在ESC细胞上清液中瘦素能刺激IL-6、IL-8的产生,不同浓度瘦素刺激IL-6、IL-8分泌量不同,各组间比较有显著性差异(P<0·05,P<0·01)。结论:瘦素是细胞因子在子宫内膜产生的一种附加调节剂,通过调节局部细胞因子而调节生殖生理过程。 Objective: To observe the effects of leptin on the secretion of IL - 6 and IL - 8 in the endometrial stromal cell ( ESC), and to evaluate leptin in the regulation of female reproductive physiology. Methods : The levels of IL - 6 and IL - 8 secreted by ESC were evaluated by ELISA when leptin were added in different concentrations. Results: The concentrations of IL - 6 and IL - 8 in the supematant of cultured ESC were increased in manner with the addition of recombinant human leptin, the levels of IL - 6 and IL - 8 were increased with leptin concentrations. When leptin added reached to 10μg/ml, IL - 6 and IL - 8 were increased significantly than those of the control group(P 〈 0. 05, P 〈 0.01 ) . Conclusion: The current findings suggest that leptin may be an additional modulator of cytokines expression in the endometrium, and leptin may contribute to the physiology and pathology processes of human reproduction by the regulation of these cytokines in the local environment.
