摘要
设计并构建了三种起始密码子ATG上下游不同序列的人GM-CSF原核表达质粒,对其核苷酸序列进行分析验证后,使它们分别在大肠杆菌中表达了人GM-CSF.结果表明:pLGM3对GM-CSF的表达量比pLGM1高约6倍,比pLGM2高约3倍.
It has designed and constructed human GM-CSF protokaryotic expression plasmids with three different upstream and downstream sequences of ATG original codon. After the three sequences have been determined, they were expressed in E. coli respectively. The results indicated that the production of GM-CSF in E. coli with PLGMI was about 6 times and 3 times higher than that of GM-CSF in E.coli with PLGM2, respectively.
