Analysis of Partial VP1 Gene of Enterovirus Type 71 Isolates from 2001 to 2004 in Shenzhen

摘要 20 enteroviruses type 71(EV71)were isolated from 410 patients with hand-foot-and-mouth disease in Shenzhen from 2001 to 2004.All of the 20 enteroviruses had been tested by RT-PCR using EV71 VP1 gene specific primers,the positive isolation rate was 4.88%.The amplicons of 341bp segment were cloned into pGEM-T and sequenced.A phylogenetic tree was constructed for comparison of these sequences with those of EV71 genotypes A,B and C using TreeView and PHYLIP software(3.6b).The sequence analysis showed that 20 EV71 strains of Shenzhen shared 81.2%-85.3% nucleotide identity with genotypes A and B.The nucleotide identities among 20 strains were 92.0%-100%.They shared 92.0%-97.9% nucleotide identity with EV71 genotype C4,and can be denominated as EV71 subgenotype C4.This research shows that part of the VP1 gene(341bp) can be used for phylogenetic analysis and pathogen surveillance. 20 enteroviruses type Shenzhen from 2001 to 2004 71(EV71)were isolated from 410 patients with hand-foot-and-mouth disease in All of the 20 enteroviruses had been tested by RT-PCR using EV71 VP1 gene specific primers, the positive isolation rate was 4.88%.
