20 enteroviruses type 71(EV71)were isolated from 410 patients with hand-foot-and-mouth disease in Shenzhen from 2001 to 2004.All of the 20 enteroviruses had been tested by RT-PCR using EV71 VP1 gene specific primers,the positive isolation rate was 4.88%.The amplicons of 341bp segment were cloned into pGEM-T and sequenced.A phylogenetic tree was constructed for comparison of these sequences with those of EV71 genotypes A,B and C using TreeView and PHYLIP software(3.6b).The sequence analysis showed that 20 EV71 strains of Shenzhen shared 81.2%-85.3% nucleotide identity with genotypes A and B.The nucleotide identities among 20 strains were 92.0%-100%.They shared 92.0%-97.9% nucleotide identity with EV71 genotype C4,and can be denominated as EV71 subgenotype C4.This research shows that part of the VP1 gene(341bp) can be used for phylogenetic analysis and pathogen surveillance.
