摘要
目的了解二硝基氯苯(DNCB)结合乙肝免疫球蛋白(HBIG)后对其抗体功能的影响。方法把HBIG(13mg/500μL PBS)分别与不同剂量(0.025,0.25,2.5mg)的DNCB(预先溶于DMSO,10mg/mL)结合,并予系列倍比稀释十次后,分别以DMSO及无菌水为两对照组,共组成五组,55个标本。以ELISA法分别检测乙肝表面抗体的效价。结果DMSO、无菌水、低剂量、中剂量与高剂量组五组的抗体阳性率分别为90.91%(10/11)、72.73%(8/11)、81.82%(9/11)、81.82%(9/11)和72.73%(8/11),其抗体阳性率以,检验无差异(P〉0.05)。经双因素方差分析(two-way analysis of variance),五组间在各稀释度的吸光值(OD450)上无差异(P〉0.05),而各稀释度处理组间则有非常显著差异(P〈0.01)。结论在一定范围内,DNCB的结合对HBIG的抗体效价没有影响,推测免疫球蛋白(抗体或单克隆抗体)可作为DNCB(或其它类似半抗原)靶向治疗的一种有效载体。
Objective To investigate the influence of dinitrochlorobenzene (DNCB) on the antibody titer of hepatitis B immune globulin (HBIG). Methods The different doses (0.025, 0.25, and 2.5mg) of DNCB (dissolved in DMSO 10mg/mL) were combined with 13mg HBIG respectively, in a volume of 500ul phosphate buffer saline (PBS). DMSO and pure water were used as 2 controls respectively. All the 5 groups were half-diluted consecutively with PBS and tested the antibody titer of HBIG with ELISA. Results The positive rates of 5 groups were 90.91% (10/11, DMSO group), 72.73% (8/11, water group), 81.82% (9/11, low dose group), 81.82% (9/11, middle dose group), and 72.73% (8/11, high dose group), respectively. There were no differences in the 5 groups in terms of positive rate (P〉0.05), nor the absorbance values (OD450) of HBIG in same dilution grade of any group with two-way anova (analysis of variance) (P〉0.05). However, there is obvious differences between different dilution grades in any group with two-way anova (P〈0.01). Conclusions It seems that DNCB has no effect on the antibody titer of HBIG at least in a certain concentration range, and the immune globulin (antibody) might be used as a carrier for DNCB (or other analog hapten) target therapy.
