茯苓、甘草、银花复方制剂对铅染毒小鼠铅、钙、锌等指标的影响 被引量:8

摘要 目的:观察茯苓、甘草、银花复方制剂对铅染毒小鼠铅、钙、锌水平的影响。方法:实验于2005-09-02/10-08在右江民族医学院重金属与氟砷毒物研究实验室进行。用60只小白鼠,数字表法随机分成5组(n=12),即葡萄糖酸锌组、葡萄糖酸锌+海尔福组、海尔福组、模型组、正常组,前4组用2g/L醋酸铅水溶液作腹腔注射染毒,1次/d,连续35d,正常组用等体积的生理盐水腹腔注射。染毒后第15天起,葡萄糖酸锌组用葡萄糖酸锌水溶液(哈药集团三精制药有限公司生产,批号:05040331)0.02118mg/只灌胃;海尔福组给予茯苓、甘草、银花复方制剂(商品名海尔福口服液,右江民族医学院海尔福研制中心提供,批号:20050618)0.12mL/只;葡萄糖酸锌+海尔福组用葡萄糖酸锌加茯苓、甘草、银花复方制剂混合后灌胃,剂量同上;模型组和正常组用等体积的蒸馏水灌胃。给药均为1次/d,至实验结束。结束时从眼球后取血,分离血清,处死动物,取大脑,冰浴下匀浆。测定血清铅、锌、砷及脑铅、脑钙水平。结果:48只小鼠进入结果分析。①脑铅:模型组高于正常组、葡萄糖酸锌组、葡萄糖酸锌+海尔福组和海尔福组眼(22.241±10.782),(5.253±0.999),(15.300±4.891),(13.460±4.132),(9.570±3.813)μg/L,P<0.01演,葡萄糖酸锌组、葡萄糖酸锌+海尔福组高于正常组(P<0.05)。②脑钙:模型组低于正常组、葡萄糖酸锌组、葡萄糖酸锌+海尔福组和海尔福组眼(0.06±0.02),(0.22±0.03),(0.19±0.01),(0.10±0.02),(0.11±0.02)mmol/L,P<0.01演,葡萄糖酸锌+海尔福组和海尔福组低于葡萄糖酸锌组(P<0.01)。③血铅:模型组高于正常组、葡萄糖酸锌组、葡萄糖酸锌+海尔福组和海尔福组眼(0.0911±0.0318),(0.0103±0.0026),(0.0709±0.0129),(0.0522±0.0518),(0.0547±0.0175)mmol/L,P<0.05或0.01演,3个用药组仍高于正常组(P<0.05)。④血锌:葡萄糖酸锌组、葡萄糖酸锌+海尔福组和海尔福组高于模型组眼(0.0968±0.0385),(0.0849±0.0103),(0.0911±0.0318),(0.0235±0.0372)mmol/L,P<0.05演。⑤血砷水平各组间尚无显著性差异。结论:茯苓、甘草、银花复方制剂有明显排铅作用,并有提高脑钙、血锌水平的功能。 AIM: To observe the effects of compound preparation of hoelen, sweet root and flos lonicerae on levels of lead, calcium and zinc of mice with lead poisoning. METHODS: The experiment was conducted in the Laboratory of Heavy Metal and Fluomsis-Arsenism of Youjiang Medical College for Nationalities from 2^nd September to 8^th October 2005. Sixty mice were randomly divided into five groups with 12 mice in each group: zinc gluconate group (ZG group), zinc gluconate + Haierfu group (ZGH group), Haierfu group, model group and normal group. Mice in the former 4 groups were poisoned with 2 g/L lead acetate water solution by abdominal injection once a day for 35 continuous days. Mice in the normal group were abdominally injected with normal saline at the same volume, Mice in the ZG group were lavaged with 0,021 zinc gluconate liquid (manufactured by Sanjing Pharmaceutical Co., Ltd, of Harbin Pharmaceutical Group with the batch number of 05040331) and 18 mg each mouse, Mice in the Haierfu group were given compound preparation (Haierfu oral liquid, provided by the Manufacture Center of Haierfu, Youjiang Medical College for Nationalities with the batch number of 20050618) and 0.12 mL for each mouse, Mice in the ZGH group were intragastrically administrated with zinc gluconate and compound preparation at the same dose, Mice in the model group and normal group were given gastric perfusion of equal-volume distilled water once a day, The blood were obtained from eyeballs at the end of experiment, and the serum was isolated, Mice were executed to obtain the brain, which was homogenated under iced bath, Levels of lead, zinc, arsenum in serum as well as the levels of lead and calcium in brain tissues were determined, RESULTS: A total of 48 mice were involved in the analysis of results,① Level of lead in brain: it was higher in the model group than that in the normal group, ZG group, ZGH group and Haierfu group [(22.241±10.782), (5.253±0.999),(15.300±4.891),(13.460±4.132),(9.570±3.813) μg/L,P 〈 0.01]. and it was higher in ZG and ZGH group than that in the normal group (P 〈 0.05).②Level of calcium in brain: it was lower in the model group than that in the normal group, ZG group, ZGH group and Halerfu group [(0.06±0.02), (0.22±0.03), (0.19±0.01), (0.10±0.02), (0.11 ±0.02) mmol/L, P 〈 0.01], and it was lower in ZGH group and Haierfu group than that in the ZG group (P 〈 0.01). ③Level of lead in serum: it was higher in the model group than that in the normal group, ZG group, ZGH group and Haierfu group [(0.091 1±0.031 8),(0.010 3±0.002 6), (0.070 9±0.012 9), (0.052 2±0.051 8),(0.054 7±0.017 5) mmol/L,P 〈 0.05 or 0.01], while that in 3 administration groups were higher than the normal group (P 〈 0.05).④Level of zinc in serum: it was higher in ZG group, ZGH group and Haierfu group than that in the model group [(0.096 8±0.038 5), (0.084 9±0.010 3), (0.091 ±0.031 8),(0.023 ±0.037 2) mmol/L,P 〈 0.05].⑤There was no significant differences in level of serum arsenum among all groups. CONCLUSION; Compound preparation of hoelen, sweet root and flos lonicerae has obvious effects of lead excretion, which can enhance the level of calcium in brain and zinc in serum.
