摘要
目的观察凋亡相关基因Survivin、Caspase-3、Caspase-9和CyclinB1在胃癌组织中的表达以及探讨Survivin与Caspase-3、Caspase-9和CyclinB1的关系。方法RT-PCR方法检测Survivin、Caspase-3、Caspase-9和CyclinB1在30例胃癌组织、10例正常胃组织标本中的表达。结果30例胃癌组织中20例SurvivinmRNA表达阳性,10例正常胃组织无SurvivinmRNA表达;30例胃癌组织和10例正常胃组织均有Caspase-3和Caspase-9以及CyclinB1的mRNA表达,但胃癌组织Caspase-3和Caspase-9及CyclinB1的mRNA表达水平明显低于正常胃组织;在20例SurvivinmRNA表达阳性的胃癌组织中,Caspase-3和CyclinB1的mRNA表达水平明显低于10例SurvivinmRNA表达阴性的胃癌组织,而Caspase-9mRNA表达水平却与10例SurvivinmRNA表达阴性的胃癌组织相差不大。结论Survivin、Caspase-3、Caspase-9和CyclinB1在胃癌组织中均存在表达,且Survivin的表达与Caspase-3和CyclinB1表达呈负相关,与Caspase-9表达不相关。
Objective To investigate expressions of apoptosis-associated genes including Survivin, Caspase-3, Caspase-9 and Cyclin-B1 in gastric cancer tissues and discuss the relationship of Survivin with Caspase-3, Ca.spase-9 and Cyclin B1. Methods RT-PCR was employed to detect the expressions of Survivin, Caspase-3, Caspase-9 and Cyclin B1 in 30 gastric cancer tissues and 10 normal gastric tissues. Results Survivin mRNA was expressed in 20 out of 30 gastric cancer tissues (66.7%) but showed no expression in 10 normal gastric tissues. Although mRNA expressions of Survivin, Caspase-3, Caspase-9 and Cyclin B1 were found in both 30 gastric cancer tissues and 10 normal gastric tissues, yet the mRNA expression level in gastric cancer tissues was significantly lower than that in normal gastric tissues. The mRNA expression levels of Caspase-3 and Cyclin Ba in 20 Survinvin-positive gastric cancer tissues were obviously lower than those in the other 10 Survinvin-negative tissues, with insignificant difference upon mRNA expression level of Caspase-9. Conclusions Survivin, Caspase-3, Caspase-9 and Cyclin B1 express in gastric cancer tissues, with Survivin expression negatively relating with expressions of Caspase-3 and Cyclin B2 but none with Caspase-9 expression.
