大豆子叶节离体再生体系优化研究 被引量:16

摘要 为获得大豆子叶节高效离体再生体系的优化方案,选用5个大豆品种的子叶节作为外植体,研究了种子萌发天数、外植体大小、不同激素浓度对大豆子叶节再生的影响。结果显示,大豆苗龄以5—7d最佳;外植体以保留全部子叶为宜;外植体的萌发和诱导均存在基因型差异,292黄豆的最佳萌发培养基为1/2MS+1mg/L6-BA,而鄂8157、湘春豆18号、湘春豆13号、湘春豆15号的最佳萌发培养基为1/2MS+2mg/L6-BA,292黄豆、鄂8157、湘春豆18号、湘春豆13号、湘春豆15号的最佳诱导培养擎组合分另0为MS+2mg/L6-BA、MS+2mg/L6-BA、MS+1mg/L6-BA、1/2MS+2mg/L6一BA+0.05mg/LIBA,MS+2Ing/L6—1;^、丛生芽诱导率分别为60%、62.5%、81.25%、77.27%和47.5%;子叶节丛生芽生根时IBA浓度以2mg/L为宜。 The cotyledonary nodes were used as explants from five excellent cultivars, soybean 292, e8157, Xiangehun 18, Xiangchun 13 and Xiangchun 15. Effects of seedling ages, explant size and hormone combinations on shoot regeneration were studied. The results showed that the germination medium 1/2 MS + 1 mg/L 6-BA was the best for soybean 292, while the medium 1/ 2 MS + 2 mg/L 6-BA was fit for the rest. The full cotyledonary is the best explant size for shoot differentiation. The best inductive mediums for soybean 292, e8157, Xiangchun 18, Xiangchun 13 and Xiangchun 15 were MS + 2 mg/L 6 - BA, MS + 2 mg/L 6 - BA, MS + 1 mg/L 6-BA, 1/2 MS + 2 mg/L 6-BA + 0.05 mg/L IBA and MS + 2 mg/L 6-BA, respectively. The shoot regeneration percentage from cotyledons of five cultivars reached 60% , 62.5% , 81.25% , 77.27% and 47.5% , respectively. Seedling age from 5 to 7 days was the best.
