摘要
目的:研究蓝舌病毒湖北株致人宫颈癌HeLa细胞死亡方式及其作用机制。方法:原位末端标记(TUNEL法)观察病毒诱导细胞凋亡情况;激光扫描共聚焦显微镜检测荧光标记的细胞核及内质网变化;萤光照度仪检测病毒诱导细胞Caspase-3/7,Caspase-8,Caspase-9活性变化。结果:凋亡染色未见明显凋亡细胞,共聚焦检测见细胞核膜不完整,染色质分散,有的凝集在核膜周围,部分染色质外溢,内质网分散,混浊不清,细胞与细胞之间分界不清;萤光照度仪检测酶活性可见Caspase-3/7,Caspase-8,Caspase-9活性均无明显变化。结论:蓝舌病毒湖北株诱导HeLa细胞胀亡而发挥其抑瘤作用。
Objective: To study the death mode of HeLa tumor cells induced by Bluetongue virus(BTV) strain HbC3 and the mechanism of induction. Methods: TUNEL staining was taken to detect the apoptosis of tumor cells induced by BTV-HbC3. The changes of the cell endoplasmic reticulum and cell nucleus treated with BTV-HbC3 were further examined with laser scanning confocal microscope. The activity of Caspase-3/7, Caspase-8 and Caspase-9 was determined by fluorescent analysis. Results: HeLa cells were sensitive to BTV-HbC3. Few apoptosis cells was found by TUNEL staining. The results of laser scanning confocal microscope showed that the nuclear membrane was discontinuous, and the chromatin in cell nucleus was dispersed partially and clumped around the nuclear membranes. Unclear periphery endoplasmic reticulums were also observed. The activity changes of Caspase-3/7, Caspase-8 and Caspase-9 were not found. Conclusion: The features of cell death in HeLa cells could be described as oncosis.
