摘要
目的:探讨转录因子T-bet/GATA-3比值在支气管哮喘免疫失衡中的意义.方法:SPF级SD大鼠随机分为对照组和哮喘组,每组12只.用逆转录-聚合酶链反应(RT-PCR)测定法和Western blot法分别检测T-bet,GATA-3 mRNA和蛋白在肺组织的表达.结果:大鼠肺组织中对照组和哮喘组T-bet mRNA表达分别为0.71±0.19,0.08±0.12;T-bet蛋白表达分别为0.72±0.22,0.18±0.06,两组比较均有显著性差异(P<0.01).大鼠肺组织中对照组和哮喘组GATA-3 mR-NA表达分别为1.06±0.25,1.56±0.28;GATA-3蛋白分别为1.04±0.44,2.25±0.74,两组比较均有显著性差异(P<0.01);对照组T-bet/GATA-3 mRNA表达量的比值0.73±0.32,明显高于哮喘组0.06±0.09(P<0.01),对照组T-bet/GATA-3蛋白表达量的比值0.75±0.25,高于哮喘组0.09±0.04(P<0.01).结论:转录因子T-bet/GATA-3 mRNA和蛋白表达量比值在哮喘中明显降低,可作为评价哮喘免疫失衡的客观指标之一.
AIM: To investigate the significance of the T-bet/ GATA-3 ratio in the immune disbalance of the asthmatic rats. METHODS: Twenty-four male SD rats were randomly divided into a control group and an asthma group ( n = 12 per group). The expressions of T-bet and GATA-3 mRNA and protein in the lungs were detected by revere transcriptase-polymerase chain reaction (RT-PCR) and Western blot respectively. RESULTS: In the control group, the expression of T-bet mRNA was 0.71± 0.19, which was significantly elevated compared with that of the asthma group(0.08±0. 12, P 〈0.01); the levels of T-bet protein expression were 0.72 ±0.22 in the control group and 0.18±0.06 in the asthma group ( P 〈 0.01 ) ; The levels of GATA-3 mRNA expression were 1.06±0.25 in the control group and 1.56± 0.28 in the asthma group (P 〈 0.01 ) ; in the asthma group, GATA-3 protein expression ( 1.04± 0.44) increased compared with that of the control group(2.25± 0.74, P 〈0.01 ) ; in the control group, the ratio of gene expression of T-bet/GATA-3 ( 0.73± 0.32 ) was significantly increased compared with that of the asthma group (0.06±0.09, P 〈0.01 ) , and there was a significant difference between the control group ( 0.75 ± 0.25 ) and the asthma group (0.09 ±0.04) in the ratio of protein expression of T-bet/GATA- 3 (P 〈 0.01 ). CONCLUSION: The mRNA and protein ratios of T-bet/GATA-3 decrease significantly in asthma, which may be one of the objective indexes to evaluate the immune disbalance in asthma.
